El concepto de nanotecnología surgió desde el año 1959, donde el Dr. Richard
Feynman habló por primera vez sobre la posibilidad de manipular átomos y moléculas,
pero no fue hasta 1970 que el Dr. Norio Taniguchi utilizó el termino nanotecnología
para describir sus investigaciones, la nanotecnología se puede definir como el estudio
y la manipulación de la materia a escala nanométrica (1-100nm). A su vez, la
nanomedicina promete innumerables soluciones y avances tecnológicos.
Para un mejor entendimiento de este proyecto fue divido en cuatro capítulos, el
primero de ellos se basó en las síntesis de nanopartículas metálicas, esta área ha
tenido un gran énfasis e interés, debido a las propiedades que estas presentan a causa
del cambio en sus características en escala nanométrica, en este proyecto se trabajó
con tres diferentes nanopartículas de oro (AuNPs), las cuales fueros las nanoesferas
de oro (AuNEs), nanopartículas de plata (AgNPs) y nanoshells de oro (AuNSs),
logrando la síntesis y caracterización, mediante dispersión de luz dinámica (DLS),
Microscopio electrónico de transmisión (MET) y espectrofotometría Uv-vis, también se
realizó un estudio de la estabilidad que estas poseen con respecto al tiempo, donde
se demostró que las AUNEs muestran una mayor estabilidad con respecto a las
AgNPs y AuNSs.
El segundo capítulo del proyecto es un estudio de los mecanismos de entrada
de las AuNEs con respecto al tiempo en células HEK-293, ya que un aspecto
importante a considerar cuando se trabaja con nanopartículas es la interacción que
existe con las células, en específico el conocer el mecanismo de absorción de las
nanopartículas. Dos procesos fundamentales a la hora de evaluar si existe efectos
citotóxicos celulares causados por las nanopartículas son la endocitosis y exocitosis,
se optó por el uso de AuNEs debido a las características que se observaron en el
capítulo I.
La tercera etapa del proyecto se basó en la encapsidación de las nanopartículas
sintetizadas en el capítulo I con la cápside del virus de planta CCMV, esto con el fin de
poder utilizar la cápside viral de los virus como nanocontenedores para las AuNPs
dándoles una mayor biocompatibilidad, durabilidad y estabilidad, para que puedan ser
utilizadas en terapias biomédicas.
La última etapa del proyecto lleva por nombre estudio del proceso de
hinchamiento de los virus Cowpea Chlorotic Mottle Virus (CCMV) y Brome Mosaic
Virus (BMV), en esta etapa del proyecto primeramente se llevó a cabo la amplificación
y purificación de los virus CCMV y BMV, posteriormente se realizó la evaluación de los
cambios del diámetro de la cápside viral, cuando se expone a diferentes condiciones
de pH y fuerza iónica (FI), estos estudios surgieron a partir de la pregunta ¿cómo es
que la cápside protege al genoma viral de los cambios de pH y FI que va encontrando
en su camino? Es decir, desde que el virus entra al hospedero y llega a la célula hay
diferentes condiciones fisicoquímicas de pH y FI, existen antecedentes que han
demostrado que los virus sufren una hinchazón, que anteriormente se creía que era
de un 10% en la cápside viral, debido a este incremento en el tamaño, surge la
hipótesis de que los extremos 5`y 3`del material genético quedan expuestos, esto daría
la posibilidad a la maquinaria celular de tomar el RNA y seguir con la replicación. Por
eso razón el CCMV y BMV se sometieron a diferentes condiciones de pH y FI, los
cuales fueron evaluados por DLS, MET y microscopia de fuerza atómica (AFM), donde
se demostró que las cápsides virales tienen un hinchamiento superior al 10% reportado
con anterioridad en la literatura.

The second chapter of the project is a study of the entry mechanisms of AuNEs with respect to time in HEK-293 cells, since an important aspect to consider when working with nanoparticles is the interaction that exists with the cells, specifically the know the absorption mechanism of nanoparticles. Two fundamental processes when evaluating whether there are cellular cytotoxic effects caused by nanoparticles are endocytosis and exocytosis. The use of AuNEs was chosen due to the characteristics observed in chapter I.
The third chapter of the project was based on the encapsidation of the nanoparticles synthesized in chapter I with the capsid of the plant virus CCMV, this to be able to use the viral capsid of the viruses as nanocontainers for the AuNPs, giving them greater biocompatibility. The last stage is called a study of the swelling process of the Cowpea Chlorotic Mottle Virus (CCMV) and Brome Mosaic Virus (BMV). For this reason, CCMV and BMV were subjected to different pH and FI conditions, which were evaluated by DLS, MET and atomic force microscopy (AFM), where it was demonstrated that the viral capsids have a swelling greater than the 10% reported with previously in the literature.
