Caracterización del péptido FLO producido en microalgas y evaluación de la actividad antimicrobiana y citotóxica en Nannochloropsis oculata

Abstract

Existe una creciente preocupación por la falta de tratamientos efectivos para infecciones causadas por bacterias resistentes, ya que la resistencia microbiana está en aumento a nivel mundial. Los PAM tienen múltiples mecanismos de acción que los vuelven efectivos contra bacterias resistentes, lo que los convierte en una opción efectiva contra infecciones causadas por estos microorganismos. El péptido antimicrobiano Flo, derivado del árbol Moringa oleifera, ha sido ampliamente estudiado por su mecanismo de acción contra bacterias. En este proyecto, se utilizaron las microalgas Scenedesmus acutus, Nannochloropsis oculata y Chlorella vulgaris para producir el péptido Flo de forma recombinante. Se demostró que la transformación genética de las microalgas con el gen flo fue exitosa, y se identificaron líneas transplastómicas en las tres cepas. La identificación y cuantificación del péptido Flo se realizó por ensayo de ELISA, en N. oculata y S. acutus. Posteriormente fue posible identificar por Western blot el péptido en N. oculata. Los ensayos antimicrobianos utilizando la proteína total soluble (PTS) de la cepa transplastómica y silvestre de N. oculata, demostraron tener actividad atribuible al péptido contra las cepas ATCC Escherichia coli, Klebsiella pneumoniae y Staphylococcus aureus, no se demostró actividad antimicrobiana para el caso de Enteroccocus faecalis. Se analizó la actividad en cepas bacterianas resistentes a antibióticos de uso clínico, donde se demostró la inhibición de Staphylococcus epidermidis, E. faecalis y K. pneumoniae, mientras que para E. coli solo se observó disminución del crecimiento bacteriano. La respuesta inflamatoria de la línea transplastómica fue analizada por el ensayo de secreción de interleucina 6 (IL-6), interleucina 10 (IL-10) y factor alfa de necrosis tumoral (TNF-α).

There is a growing concern about the lack of effective treatments for infections caused by resistant bacteria, as microbial resistance is on the rise worldwide. Antimicrobial peptides (AMPs) have multiple mechanisms of action that make them effective against resistant bacteria, making them a viable option for combating infections caused by these microorganisms. The antimicrobial peptide Flo, derived from the Moringa oleifera tree, has been extensively studied for its mechanism of action against bacteria. In this project, the microalgae Scenedesmus acutus, Nannochloropsis oculata, and Chlorella vulgaris were used to produce recombinant Flo peptide. Successful genetic transformation of the microalgae with the flo gene was demonstrated, and transplastomic lines were identified in all three strains. Identification and quantification of the Flo peptide were conducted using an ELISA assay, in N. oculata and S. acutus, respectively. Subsequently, the peptide was identified in N. oculata through Western blot analysis. Antimicrobial assays using the soluble total protein (PTS) from the transplastomic and wild strains of N. oculata showed activity attributed to the peptide against ATCC strains of Escherichia coli, Klebsiella pneumoniae, and Staphylococcus aureus; however, no antimicrobial activity was demonstrated against Enterococcus faecalis. The activity against clinically used antibiotic-resistant bacterial strains was analyzed, revealing inhibition of Staphylococcus epidermidis, E. faecalis, and K. pneumoniae, while only a reduction in bacterial growth was observed for E. coli. The inflammatory response of the transplastomic line was analyzed using interleukin 6 (IL-6), interleukin 10 (IL-10), and tumor necrosis factor-alpha (TNF-α).
